Introduction to Fragment Analyzer Service
The Molecular Biology Core Lab (MBCL) at UMass Chan Medical School offers a Fragment Analyzer Service, utilizing the Advanced Analytical (Agilent) capillary electrophoresis system. This service evaluates samples for concentration and molecular weight/size, providing results in a graphic format similar to a Bioanalyzer, including size, concentration, molarity, and RIN values.
Available Assays and Submission Requirements
The Fragment Analyzer Service offers various assays, including:
- small RNA
- mRNA
- total RNA
- DNA
- large fragment DNA
- genomic DNA
There is no minimum or maximum number of samples required, and only 3 µl per sample is needed. The resolution can be as low as 3 bp, and DNA fragment sizing ranges from 25 bp to 48 Kb, depending on the assay.
Submitting Samples
To submit samples, please email MBCL@umassmed.edu for a ticket or more information. Samples can be dropped off at designated locations, and off-campus customers can ship samples to the Rose-Gordon Bldg. Rm. 141, 222 Maple Ave, Shrewsbury, MA, 01545.
Helpful Tips for Sample Preparation
- Plan ahead: Results will be ready by the end of the next business day after the scheduled run.
- Sample suspension: Samples should be suspended in water, as contaminants can create problems with the run.
- Assay selection: RNA, dsDNA, and ssDNA can be detected by the machine, and will be visible if they are in your sample, regardless of the selected assay.
- Concentration estimation: Provide an estimate of the sample concentration to ensure the chosen assay matches the sample concentrations.
- Optimal concentration ranges: Submit samples at concentrations between 0.5 and 1 ng/μl for High Sensitivity DNA Assays, and at the upper end of the concentration range for High Sensitivity RNA assays.
- NanoDrop measurements: Be aware that NanoDrop readings may be higher than Fragment Analyzer results due to contamination detection.
- Sample naming: Sample names are imported from the electronic ticket, so please submit the excel sheet and paper copy.
Fragment Analyzer Pricing and Payment Policy
Please email MBCL@umassmed.edu for a quote for your desired services. Payment is the responsibility of the user submitting the sample and should be rendered in a timely fashion.
